Working Principle
|
This method is particularly used to determine the amount of fructose in sperm samples because fructose is the energy source of spermatozoites. Fructose concentration is an important indicator for assessing the function and motility of spermatozoites. Low fructose levels may be a sign of dysfunctions in the seminal vesicles or other reproductive system problems. It is an assay for the direct determination of fructose in spermatozoites. The study uses semen from which proteins have been removed by precipitation. It is based on the principle that fructose turns red when heated with hydrochloric acid against resorcin. The fructose level in healthy semen is around 315 mg/dl. Decreases below this figure necessitate the investigation of a pathological event. |

Wear protective gloves/protective clothing/eye protection/face protection. IN CASE OF CONTACT WITH EYES: Rinse carefully with water for several minutes. Remove contact lenses if fitted and easy to do so. Continue rinsing. |

Application
|
The semen to be examined is allowed to liquefy. Take a test tube. 0.1 ml semen, 2.9 ml distilled water, 0.5 ml A-Barium Hydroxide Solution and 0.5 ml B-Zinc Sulphate Solution are put into the tube respectively. Mix and leave for 3-5 minutes. Centrifuge at high speed. The upper part is separated with a pipette. This prepared solution is the “Test Solution”.
Working Standard:
It should be prepared fresh.
0.1 ml of C-Fructose Stock Standard Solution is taken into a tube. Add 9.9 ml of distilled water. It is mixed. This solution is the “Working Standard” and corresponds to 5% mg (5 mg/dl) fructose.
1) Take 3 test tubes.
2) Mix the tubes properly.
3) Heat the tubes together in a boiling water bath at 90 °C for 10 minutes.
4) Cool all together with running water.
5) Placed in cuvettes. Read at 490 nm against the blind. |
