Purpose |

Working Principle
|
Elastic dyes bind to elastin proteins, making elastic fibers appear darker. This helps to highlight the elastic fibers in tissue sections. A mixture of picro acid and acidic fuchsin stains collagen fibers red and other connective tissue components yellow. This different staining allows collagen and elastic fibers to be easily distinguished from each other. This staining method provides important information in textural and pathological examinations by clarifying elastic and collagen fibers. The appearance of elastic fibers in dark purple/black and collagen fibers in red allows these structures to be easily distinguished under light microscopy. |

Hazard Highly flammable liquid and vapor. Harmful if swallowed. May cause damage to organs. Causes allergic skin reactions. Causes severe eye and skin irritation. Keep away from heat/sparks/flame/hot surfaces. Do not smoke. Keep container tightly closed. Use only non-flammable tools. IN CASE OF CONTACT WITH EYES: Rinse carefully with water for several minutes. Remove contact lenses if fitted and easy to do so. Continue rinsing. INHALATION: Remove the affected person to fresh air and keep in a comfortable position so that he/she can breathe easily. In case of exposure or unwellness: Call NATIONAL POISON ADVICE CENTER PHONE 114 or doctor/physician. Dispose of contents/container in accordance with national regulations. |

B) Weigert Resorcin Fuchsin Stain: Fucsin, Resorcinol, Ethanol 96%, Ferric chloride
C) Weigert Ferrous Hematoxylin-A solution: Ethanol 96%, Methanol, Hematoxylin
D) Weigert Ferrous Hematoxylin-B solution: Iron (III) chloride, Hydrochloric acid 37%
E) VanGieson's Picric Acid Fuchsin solution: Picric acid Fuksin
F) Alcoholic Fixative: Acetic acid, Methanol |

Application
|
1.Place the tissue sections in distilled water and wait for 2 minutes.
2.Remove from distilled water and drop 5 drops of ‘A-Periodic acid solution 0.5%’ onto the tissue section.
3.Wait for 5 minutes and wash with distilled water.
4.Leave covered in the working solution overnight.
To obtain results in a shorter time in this step;
Add 20 drops of ‘F-Alcoholic fixative’ to the incubation box. Immerse the preparation and add 10 drops of ‘B-Weigert resorcin fuchsin dye’. Close the incubation box and incubate for 30 minutes.
5. Shake in ‘F- Alcoholic fixative’.
6.Wash under running distilled water for 10 minutes.
7.5 drops of ‘C- Weigert Ferrous Hematoxylin A solution’ and 5 drops of ‘D- Weigert Ferrous Hematoxylin B solution’ are added and waited for 10 minutes.
8.Wash under running water for 10 minutes.
9. Stain for 5 minutes in 10 drops of ‘E- VanGieson's Picric Acid Fuchsin Solution’.
10. Shake rapidly in distilled water.
11.Dehydrated, cleared and sealed. |
