Purpose Used for light microscopic visualization of acid mucopolysaccharides, glycogen, glycoprotein, mucoprotein, high molecular weight carbohydrates in tissue sections. Specially designed for use in formalin-fixed paraffin block tissue sections or cytologic preparations for routine tissue follow-up.
Working Principle Alcyan blue is a water-soluble polyvalent basic dye. Its blue staining property comes from the copper in the molecule. At acid pH formed by acetic acid, a) sulfated acid mucopolysaccharides, b) carboxylated acid mucopolysaccharides, c) sulfated sialomucins (glycoproteins), d) carboxylated sialomucins (glycoproteins) stain with alcyan blue. Periodic acid selectively oxidizes 1,2-glycolic, primary amine, secondary amine and hydroxy ketonic groups. High molecular weight carbohydrates carry these groups. As a result of oxidation, the ends of some carbon atoms in the structure become free. It retains leucofucsin dye (sulfurized fucsin dye) and prevents it from dissolving in water. The parts where the dye is retained appear magenta red.
Warnings, Precautions, Recommendations Caution. Irritant. Toxic. May cause cancer. In contact with acids, releases toxic gas. Prolonged or repeated exposure may cause allergic reactions in some sensitive individuals. Irritating to eyes and skin, releases toxic gas on contact with acids. Wear protective gloves/protective clothing/eye protection/face protection. IN CASE OF CONTACT WITH SKIN: Wash with plenty of soap and water. In case of inhalation: Shortness of breath, mucosal irritation, cough, headache in case of prolonged exposure. In case of skin irritation: Get medical advice/attention. In case of ingestion: Consult a doctor.

G) Mayer Hemalum Solution: Ethanol 96%, Hematoxylin, Glycerol, Aluminum ammonium sulfate

Accessories
Application 1. Cover the tissue section with 10 drops of “A- Alcyan Blue pH: 2.5 solution”. After 30 minutes, remove the reagent by shaking the slide.
2. Cover with 10 drops of “B- Borate Buffer Solution” without washing with water. After 10 minutes, shake the slide to remove the reagent.
3. Wash with distilled water for 2 minutes.
4. Cover the tissue section with 10 drops of “C- Periodic Acid Solution”. After 10 minutes, shake the slide to remove the reagent.
5. Wash with distilled water for 2 minutes.
6. Cover with 10 drops of “D- Schiff's Separator McManus”. After 20 minutes, shake the slide to remove the reagent.
7. Wash with distilled water for 2 minutes.
8. Cover with 10 drops of “E- Sodium Metabisulphite Solution”. After 2 minutes, shake the slide to remove the reagent.
9. Cover with 10 drops of “F- Fixing Solution” without washing with water. After 2 minutes, shake the slide to remove the reagent. Shorten or remove this step for significant PAS positive reaction.
10. Wash with distilled water for 2 minutes.
11. Cover with 10 drops of “G- Mayer Hemalum Solution”. After 3 minutes, shake the slide to remove the reagent.
12. Wash with distilled water for 5 minutes.
13. Dehydrate, clean, cover with synthetic resin and examine under a microscope.
