Working Principle
|
EDTA buffer solution (x10) is a widely used chemical solution in laboratories, often preferred for its chelation (ion binding) properties. EDTA (ethylenediaminetetraacetic acid) plays an important role in biochemical experiments and analysis by binding metal ions. It is used for the recovery of antigens in tissue sections and during staining processes. It improves staining results by eliminating the effect of metal ions. It is used to inhibit the adhesion of cells and to suspend cells. This allows temporary separation of cells in cell culture experiments. Inhibits enzymatic activities caused by metal ions in protein samples. This is especially important during purification of proteins and enzymes. EDTA is used to prevent the degradation of nucleotides. It inhibits nuclease activity by binding metal ions and thus ensures the protection of DNA and RNA. |

Preparation of the Solution (Dilution)
The term “x10” indicates that the solution is 10 times concentrated.
It must be diluted with distilled water at a ratio of 1:10 before use.
For example: You can prepare the solution by mixing 100 mL of x10 EDTA solution with 900 mL of distilled water.
Antigen Recovery (Heat-Induced Epitope Retrieval - HIER)
The diluted (x1) EDTA buffer solution is placed in a heat-resistant container.
The slides are placed in this solution.
The heating method is applied for antigen retrieval:
A water bath, microwave, pressure cooker, or pressure system can be used.
Typical procedure: Bring to boiling point in the microwave and heat for 15–20 minutes.
After heating, allow to cool at room temperature for 20–30 minutes.
Washing Procedure
Once the slides have reached room temperature, they are washed with distilled water or PBS and proceed to the next step (blocking or antibody incubation).
Notes
The antigen retrieval process should be optimized according to the tissue and the antibody to be used.
The heating time and method should be carefully adjusted so as not to damage the tissue structure. |
