Working Principle
|
PAS+AB Dye is a combination of Alcian Blue (AB) and PAS (periodic acid-Schiff) dye solution. Alcian, also known as alcian blue, is a kind of copper peptide cyanine dye, which is alkaline and belongs to cationic dye. Alcian blue with acid groups is used to color tissues with anionic groups (such as carboxyl group), making acidic mucopolysaccharides in the cytoplasm appear blue, while PAS dye solution is used for neutral samples.
This staining for glycogen PAS staining, also known as periodate Schiff stain, is mainly used in histomorphology to detect glycogen and polysaccharide substances in tissues. The basic principle is to oxidize hydroxyl groups on two adjacent carbon atoms in carbohydrate materials to aldehyde groups. The oxidant periodic acid and then Schiff reagent react with the aldehyde groups to produce purple red substances. Acid glycoprotein and neutral glycoprotein can be distinguished by the combination of Alcian blue and PAS stain.
After staining the specimen, acidic mucus substances are observed in blue, neutral mucus substances in red, and mixed mucus substances in blue-violet. |
Warnings, Precautions, Recommendations

Caution Toxic. Causes severe eye and skin irritation. In case of inhalation: Prolonged exposure may cause shortness of breath, mucosal irritation, cough, headache. If swallowed: A doctor should be consulted. Suspected to cause cancer. Wear protective gloves/ protective clothing/ eye protection/ face protection. In contact with skin: Immediately remove all contaminated clothing. Dry skin with water/ shower. In case of eye contact: Dry carefully with water for a few minutes. Remove contact lenses if worn and easy to do so. Continue rinsing. In case of exposure or interaction: Get medical advice/attention. Keep under lock and key. |

Application
|
1. Deparaffinize sections and hydrate in distilled water if necessary.
2. Cover the tissue section with approximately 5-10 drops of “A- Acetic Acid Solution, 3%” for 2 minutes.
3. Pour off excess ''A-3% Acetic Acid Solution'' and do not rinse.
4. Cover the tissue section with 10 drops of “B- Alcyan Blue Solution pH: 2.5”. After 30 minutes, shake the slide to remove the reagent.
5. Wash with distilled water for 2 minutes.
6. Cover the tissue section with 10 drops of “C-Periodic Acid Solution”. After 10 minutes, shake the slide to remove the reagent.
7. Wash with distilled water for 2 minutes.
8. Cover with 10 drops of “D- Schiff's Separator McManus”. After 20 minutes, shake the slide to remove the reagent.
9. Wash with distilled water for 2 minutes.
10. Cover with 10 drops of “E- Mayer Hemalum Solution”. After 3 minutes, shake the slide to remove the reagent.
11. Wash with distilled water for 5 minutes.
12 Cover with 10 drops of “F- Blueing solution”. After 30 seconds, shake the slide to remove the reagent.
13. Examine under the microscope. |
