Purpose |
It is used under the microscope to demonstrate acid-fast bacteria, especially Mycobacterium tuberculosis. |
Working Principle
|
A staining technique used to visualize acid-fast bacilli using fluorescence microscopy. Acid-fast organisms exhibit a reddish-yellow fluorescence color. Although the auramine-rhodamine stain is not as specific as the Ziehl-Neelsen stain for acid-fast organisms (e.g., Mycobacterium tuberculosis or Nocardia ), it gives more rapid results and is therefore often used as a screening tool. |

Danger. Highly flammable liquid and vapour, Keep away from heat/sparks/flames/hot surfaces, No smoking. May cause severe skin burns and eye damage. May cause drowsiness or dizziness. May cause genetic defects. Wear personal protective equipment. IF SWALLOWED: Rinse mouth. DO NOT induce vomiting. IF ON SKIN (or hair): Remove/take off immediately all contaminated clothing. Rinse skin with water/shower. IF IN EYES: Rinse cautiously with water for several minutes. Remove contact lenses if present and easy to do. Continue rinsing. Store locked up. |

A) Auramine Rhodamine Solution: Auramine o, Rhodamine B, Glycerol, Phenol, Propanol.
B) Acidic Bleach Solution: Propanol, Hydrochloric acid.
C) Potassium Permanganate Solution: Potassium permanganate. |

Application
|
1. Cover with “A- Auramine Rhodamine Solution”. Shake off after 15 minutes.
2. Wash with distilled water.
3. Cover with “B- Acidic Bleach Solution”. Shake off after 1 minute. You can change the reagent several times during this period.
4. Wash with distilled water.
5. Cover with “C- Potassium Permanganate Solution”. Shake off the reagent after 3 minutes.
6. Wash with running distilled water.
7. Examine under a microscope. |
