Purpose

It is used to distinguish pathogenic bacteria, particularly Koch's bacillus and Hansen's bacillus, by staining them in histological sections, sputum, and culture smears. It is a special staining kit used to stain weakly acid-fast bacteria, such as Mycobacterium leprae (the causative agent of leprosy), in histological sections.

Working Principle

Mycobacterium, Nocardia, and Corynebacterium do not contain teichoic acid in their cell walls. Approximately 35% of the cell wall consists of lipids, particularly mycolic acid. It is hypothesized that this characteristic of the cell wall increases its resistance to acids and alcohols. These bacteria do not easily release the stain they receive; that is, they retain the fuchsin stain. Since the wall of bacilli is resistant to acidic alcohol, the dye remains in the bacillus. The Fite method performs a gentler decolorization compared to classical acid-resistant staining. This increases the chances of staining weak acid-resistant microorganisms.

Application

SAMPLE AND PREPARATION:
1. Body fluids may be subjected to known homogenization processes prior to examination. Working with samples to be examined for Mycobacterium tuberculosis carries a serious risk of contamination. Therefore, it is preferable to stain and examine samples that have been treated with hypochlorite. Processing for less than 15 minutes does not cause structural damage to the bacilli and does not result in differences in staining quality.
2. Fixation: Preferably, they are fixed for 2 hours at 65-75°C on heated plates. Fixation by flame heating is widely used because it is practical.
3. Staining method: Covering for smears, dipping for tissue sections.

STAINING:
1. Cover the preparation with “A-Periodic Acid Solution”. Wait 10 minutes.
2. Wash with distilled water.
3. Cover the preparation with “B- Carbol Fuchsin Solution ZN”. Wait for 30 minutes.
4. Wash with distilled water. Dry with filter paper.
5. Cover the preparation with “C- Acid Alcohol Solution”. Wait for 1 minute.
6. Rinse with tap water for 3 minutes.
7. Cover the specimen with “D- Methylene Blue Solution.” Let it sit for 1 minute.
8. Wash with distilled water.
9. Dry, clean, cover with synthetic resin, and examine under a microscope.
