| Purpose |
This kit is used for the differentiation of Gram-positive and Gram-negative bacteria by light microscopy. |
| Working Principle
|
The peptidoglycan structure of Gram-negative bacteria is thinner than that of Gram-positive bacteria between the outer membrane and the inner membrane. In Gram-positive bacteria, the thick peptidoglycan has the property of retaining crystal violet. The cell wall permeability of Gram-positive bacteria is lower. Therefore, they do not release dyes during decolorization with denaturing alcohol. In Gram-negative bacteria, alcohol dissolves the lipids in the outer membrane, pores open, and crystal violet disperses. Thus, Gram-negative bacteria are subsequently stained with fuchsin added to the medium. |

Hazard. Highly flammable liquid and vapor. It causes severe eye irritation. Suspected to cause cancer. Keep away from heat/ sparks/ flames/ hot surfaces. Wear protective gloves/ protective clothing/ eye protection/ face protection. In contact with skin : Immediately remove all contaminated clothing. Rinse skin with water/ shower. In case of eye contact : Rinse cautiously with water for several minutes. Remove contact lenses if fitted and easy to do so. Continue rinsing. IF exposed or concerned: Get medical advice/attention : Get medical advice/attention. Keep under lock and key. |

A) Gram Crystal Violet Solution: Crystal violet, ethanol, ammonium oxalate and Stabilizer.
B) Gram Denatured Alcohol: Ethanol 96%, methanol, 2-propanol and stabilizer.
C) Gram Lugol Solution: Iodine, Potassium Iodide,96% Ethanol and Stabilizer.
D) Gram Fuchsin Solution: Fuchsin, phenol and 96% ethanol. |

| Application
|
1. Cover the smear with A – Gram Crystal Violet Solution. Allow you to stand for 5 minutes, then drain off.
2. Rinse gently with distilled water.
3. Cover the smear with C – Gram Lugol’s Solution. Allow you to stand for 1 minute, then drain off.
4. Rinse gently with distilled water.
5. Cover the smear with B – Gram Denatured Alcohol Solution for 5–15 seconds. Immediately rinse with distilled water to stop decolorization.
6. Rinse gently with distilled water.
7. Cover the smear with D – Gram Fuchsin Solution. Allow you to stand for 1 minute, then rinse with distilled water.
8. Rinse gently with distilled water.
9. Air dry at room temperature. Examine under a microscope.
