Purpose It is usually used for staining blood cells.
Working Principle Giemsa stain is a differential stain and contains a mixture of azure, methylene blue and eosin stain. It is specific for the phosphate groups of DNA and binds itself where there is a high amount of adenine-thymine bonding. Azur and eosin are acidic dyes that variably stain the basic components of cells such as cytoplasm, granules, etc. Methylene blue, on the other hand, acts as a basic stain for acidic components, especially the cell nucleus.

Application Staining of Smears
1. Preparation of Dye Solution (Dilute Giemsa Solution):
▪ Take 5 ml of GBL-2408 "Giemsa Stain; Concentrate" into the staining container.
Add 45 ml of distilled water and mix well.
2. Staining with Giemsa Solution:
▪ Cover the preparation with methanol and fix for 5 minutes.
Immerse the preparation in the prepared staining solution.
After 15-30 minutes, remove the slide and shake to remove the reagent.
Wash with distilled water. Dry it.
Examine under a microscope.
