Purpose Clarke Fixative is an alcohol-based coagulant fixative used in histology and cytology laboratories for rapid tissue and cell fixation. It is particularly suitable for cytological preparations, smear samples, and thin tissue sections.
Working Principle Clarke fixative is usually composed of a mixture of ethanol and acetic acid. Alcohol rapidly stabilizes cell structure by coagulating cellular proteins and halting enzymatic degradation. Acetic acid is particularly effective on nuclear structures; it swells chromatin, contributing to the clearer visibility of nuclear details. This combination increases the contrast between the cytoplasm and the nucleus, allowing cellular details to be seen more clearly under microscopic examination.
Warnings, Precautions, Recommendations Danger. May cause serious eye damage and skin irritation. May cause respiratory tract irritation. The product contains a high percentage of flammable alcohol; its vapors may form explosive mixtures with air. Keep away from heat, sparks, open flames, and hot surfaces. Avoid breathing in the product's vapors or mist; the work area should be well ventilated. Chemical-resistant protective gloves, appropriate protective clothing, eye protection, and face protection equipment should be used during use. If the product comes into contact with the skin, contaminated clothing should be removed and the affected area washed thoroughly with water. If inhaled, move the person to fresh air and keep them in a position where they can breathe comfortably; seek medical attention if symptoms persist. In case of eye contact, immediately rinse eyes thoroughly with plenty of water for at least 15 minutes; if contact lenses are present and can be easily removed, do so and continue rinsing; seek medical attention if irritation persists. If swallowed, do not induce vomiting and seek medical attention immediately. In cases of exposure or suspected exposure, seek medical advice and show the product label or Safety Data Sheet to the healthcare professional. Due to its flammable content, precautions should be taken against static electricity and containers should be kept tightly closed.
