Regulation
Purpose It is used to detect acid-resistant bacteria by staining.
Working Principle Mycobacterium, nocardia and corynebacterium do not have taychoic acid in their walls. About 35% of the wall is composed of lipids and especially mycolic acid. It is hypothesised that this feature of the wall increases the resistance of the bacterial wall against acid and alcohol. They do not easily release the dye they have taken up, i.e. they retain the Fuchsin stain. Since the wall of the bacilli is resistant to acidic alcohol, the stain remains in the bacilli.

Application PREPARATION OF THE SMEAR:
1. A new, clean and scratch-free slide is taken.
2. The slides are cleaned by immersion in ethanol before use.
3. A portion of the sample is transferred to the slide using a pipette or pipette. For concentrated samples, the precipitate obtained from the concentration process is used. For non-concentrated specimens, bloody or necrotic material, if available, is used for smear preparation.
4. The sample is spread on the slide over an area approximately the size of a coin (1.5 x 1.5 cm). No more than one smear is made on the slide.
5. If smears are prepared from dense material, the two slides are pressed, pulled apart. This is done several times to produce an acceptable smear.
6. The core is thoroughly flamed before reuse. The pipette is disposed of in disinfectant or biohazard-marked containers and a new pipette is used for each sample.
7. The smear is air-dried in a biological safety cabinet.
STAINING:
1. Cover the smear with "A-Carbol Fuchsin Solution FK". After 2 minutes, remove the reagent by shaking.
2. Wash with distilled water.
3. Wash the smear with "B-Acid Alcohol solution". Discolour until the liquid flows colourless from the slide.
4. Wash with distilled water. Drain excess water from the preparation.
5. Cover the smear with "C-Methylene Blue Solution". After 30 seconds, shake the slide to remove the reagent.
6. Wash thoroughly with distilled water. Drain excess water from the preparation.
7. Dry the smear in air, preferably on a heated plate. Do not dry with blotting paper.
8. Examine the smear with a 100 immersion objective.
