Purpose

It is used in histological sections, sputum and culture smears; in the examination of pathogenic mycobacteria (especially Koch's bacillus) by staining with light microscopy.

Working Principle

The cell wall of mycobacteria contains high lipid concentrations which make them waxy, hydrophobic and impermeable to routine stains such as Gram Stain. They are also resistant to acid and alcohol and are designated as acid-fast bacilli (AFB) or acid-alcohol-resistant bacilli (AAFB).

Wear protective gloves/protective clothing/eye protection/face protection. Do not breathe dust/fume/gas/mist/vapours/spray. Use only outdoors or in a well-ventilated area. IF SWALLOWED: If you feel unwell, call the NATIONAL POISON CONSULTATION CENTER ON 114 or a doctor/physician. IF ON SKIN (or hair): Remove/take off immediately all contaminated clothing. Rinse skin with water/shower. IF IN EYES: Rinse cautiously with water for several minutes. Remove contact lenses, if present and easy to do. Continue rinsing. IF exposed or exposed: Get medical advice/attention. Store locked up.

Ingredients

A) Periodic Acid Solution: Periodic acid.
B) Carbol Fuchsin Solution ZN: Fuchsin, Phenol, Ethanol
C) Acid Alcohol Solution: Ethanol, Methanol, Hydrochloric acid.
D) Mayer Hemalum Solution: Hematoxylin, Ammonium aluminum, Sodium iodate, Glycerol. Triton X-100

Application

Sample and Preparation:
•Body fluids can be subjected to known homogenization processes before being examined.
•Working with samples that will be examined for Mycobacterium tuberculosis carries a serious risk of contamination. For this reason, it should be preferred to examine samples that have been treated with hypochlorite by staining.
The process, which takes less than 15 minutes, does not cause structural deterioration in the bacilli and does not cause differences in staining quality.
Staining method: Covering in smears, dipping in tissue sections.
Application of Staining Method:
•Rinse the preparation with distilled water.
•Cover the preparation with 10 drops of “A-Periodic Acid Solution”. Wait for 10 minutes.
When the time is up, rinse the preparation with distilled water.
•Cover the preparation with 10 drops of “B-Carbol Fuchsin Solution ZN”. Wait for 30 minutes.
When the time is up, wash with distilled water. Dry the slide with filter paper.
•Cover the preparation with 10 drops of “C- Acid Alcohol Solution”. Wait for 1 minute.
•Rinse under tap water for 3 minutes.
•Cover the preparation with 10 drops of “D- Mayer Hemalum Solution”. Wait for 2 minutes.
•Rinse with distilled water. Blue under running tap water for 5 minutes.
•Dehydrate by passing through various alcohols or denatured alcohol substitutes.
•Clean by passing through xylene or substitutes.
•Cover with synthetic resin.
Examine under a microscope.
NOTE:
To increase the distinctiveness in staining results, one or more of the following applications should be preferred:
1) Staining times should be reduced or increased.
2) Water washing times should be extended or shortened.
3) The times of procedures performed with absolute alcohol and xylene should be extended.
