Working Principle
This method targets the enzyme acid phosphatase, which is found in the cell membranes of leukocytes. Acid phosphatase is an enzyme found on the inner surface of the cell membrane and is also present in leukocytes. The enzyme acid phosphatase hydrolyzes the phosphate molecules in the staining solution. As a result of this reaction, phosphate residues are formed in the cell membranes of cells. Naphthol AS, which is released by phosphatase activity in acidic medium, forms a colored complex as a result of diazolation with fast garnet GBC dyestuff.

Application
1.Immerse the preparation in a container of “Fixative solution” brought to room temperature. After 30 seconds, shake the slide to remove the reagent. Do not allow the preparation to dry.
2.Wash with distilled water for 1 minute. Dry briefly (less than 15 seconds) or remove wetness with absorbent paper.
3.Immerse the preparations in containers with “AP incubation solution” and “TRAP incubation solution”. Incubate at 18-26 degrees C for 60 minutes, protected from light. Shake the slides out at the end of the time.
4.Wash with distilled water for 2 minutes. Dry briefly (less than 15 seconds) or remove wetness with absorbent paper.
5.Cover the preparation with 10 drops of “G- Hematoxylin solution Gill-3”. After 2 minutes, shake the slide to remove the reagent.
6.Wash several times with distilled water.
7.Air dry.
