Purpose |
It is used in the examination of acid mucopolysaccharides in tissue sections by light microscopy. |
Working Principle
|
Alcian Blue dye interacts with acidic mucopolysaccharides in a unique way. These mucopolysaccharides are found especially in mucosa and some other tissues. After the dye interacts with acidic mucopolysaccharides, it forms a complex with these compounds and this complex is blue. This blue color, which is concentrated in mucosal areas, provides the mucosa to be highlighted.
Alcian blue is a water-soluble polyvalent basic dye. The blue staining property comes from the copper in the molecule. In the acidic pH created by acetic acid,
a)sulfated acid mucopolysaccharides
b)carboxylated acid mucopolysaccharides
c)sulfated sialomucins (glycoproteins)
d)carboxylated sialomucins (glycoproteins) are stained with Alcian blue. |

Application
|
1. Cover the tissue section with 10 drops of “A- Alcian Blue pH:2.5 Solution”. After 30 minutes, shake the slide to remove the reagent.
2. Before washing with water, cover with 10 drops of “B- Borate Buffer Solution”. After 10 minutes, shake the slide to remove the reagent.
3. Wash with distilled water for 3 minutes.
4. Cover with 10 drops of “C- Modified Mayer Hemalum Solution”. After 5 minutes, shake the slide to remove the reagent.
5. Wash with distilled water for 2 minutes.
6. Dehydrate, clear, cover with xylene-based coverslip medium and examine under a microscope. |
