Used for microscopic examination of Gram positive (+) and Gram negative (-) bacteria. |
Working Principle
|
Gram Positive/Negative status is a characteristic of the cell wall (peptidoglycan) structure of bacteria. The peptidoglycan structure of Gram Negative bacteria is thinner between the outer membrane and the inner membrane than Gram Positive bacteria. In Gram positive bacteria, the thick peptidoglycan is capable of retaining Hucker crystal violet. Burke's iodine solution allows the dye to be retained. Gram positive bacteria have less cell wall permeability. Therefore, they do not release dyes during decolorization with acetone denatured alcohol. In Gram-negative bacteria, alcohol dissolves the lipids in the outer membrane, the pores open and the Hucker crystal violet disperses. Gram-negative bacteria are then stained with safranin added to the medium. |

Keep away from heat/sparks/flame/hot surfaces. Wear protective gloves/protective clothing/eye protection/face protection. IN CONTACT WITH SKIN (or hair): Immediately remove all contaminated clothing. Dry skin with water/shower. IN CASE OF CONTACT WITH EYES: Rinse carefully with water for several minutes. Remove contact lenses if fitted and easy to do so. Continue rinsing. In case of exposure or interaction IF: Get medical advice/attention. Keep under lock and key. |

Application
|
1. Cover the preparation with “A-Hucker Crystal Violet Solution”. After 1 minute, shake off.
2. Wash with distilled water for 15 seconds.
3. Cover the preparation with “B- Burke's Iodine Solution”. After 1 minute, shake off.
4. Wash with distilled water for 15 seconds.
5. Cover the preparation with “C- Denatured Alcohol Solution with Acetone” After 10 seconds, shake the slide to remove the reagent.
6. Repeat this procedure one more time.
7. Wash with distilled water for 15 seconds.
8. Cover the preparation with “D- Safranin Solution”. After 1 minute, shake the slide to remove the reagent.
9. Wash with distilled water for 15 seconds.
10. Air dry, examine under a microscope. |
