| Purpose |
It is used to stain connective and support tissues and examine them with light microscopy. |
| Working Principle
|
It is specially produced to work in paraffin block tissue sections or cytological preparations that are fixed in formalin and routine tissue follow-up. Three dyes are used to selectively stain muscle, collagen fibers, fibrin and erythrocytes. The general rule in trichrome staining is that less porous tissues are colored with the smallest dye molecule. The tissue is first stained with acid dye. At this stage, the dye binds with acidophilic tissue components. When it is then treated with phospho acids, the less permeable components retain the red, while the red is pulled out of the collagen. It also causes a bond with the collagen to bind with aniline blue. |

Danger. Highly flammable liquid and vapour. Causes skin irritation. Toxic if swallowed, in contact with skin or if inhaled. Harmful if swallowed. Causes damage to organs. IF ON SKIN (or hair): Remove/Take off immediately all contaminated clothing. Rinse skin with water/shower Keep away from heat/sparks/flames/hot surfaces. - No smoking. Wear protective gloves/protective clothing/eye protection/face protection. In case of exposure: Call the NATIONAL POISON CONSULTATION CENTER ON 114 or doctor/physician. Store in a well-ventilated area. Keep container tightly closed. Store locked up. |

A) Weigert's Iron Hematoxylin-A Solution: Ethanol 96%, methanol, hematoxylin.
B) Weigert's Iron Hematoxylin-B Solution: Iron (III) chloride, hydrochloric acid 37%.
C) Picric Acid Alcoholic Solution: Ethanol 96%, picric acid.
D) Poppy Red Fuchsin Solution: Acid fuchsin, Biebrich scarlet, glacial acetic acid, Triton X-100.
E) Phosphomolybdic Acid Solution: Phosphomolybdic acid.
F) Masson's Aniline Blue Solution: Aniline blue, glacial acetic acid |

| Application
|
1. Cover the tissue section with 5 drops of A- Weigert iron hematoxylin and 5 drops of B- Weigert iron hematoxylin. Mix the two stains by blowing and small oscillations. After 10 minutes, shake the slide to remove the reagent from the preparation. Dry the preparation at room temperature for 30-60 minutes.
2. Wash with distilled water for 3-4 seconds.
3. Cover the tissue section with 10 drops of C- Picric acid alcoholic solution. After 4 minutes, shake the slide to remove the reagent from the preparation.
4. Wash with distilled water for 3-4 seconds.
5. Cover the tissue section with 10 drops of D- Poppy red fuchsin solution. After 4 minutes, shake the slide to remove the reagent from the preparation.
6. Wash with distilled water for 3-4 seconds.
7. Cover the tissue section with 10 drops of E- Phosphomolybdic acid solution. After 10 minutes, shake the slide to remove the reagent from the preparation.
8. Dry the preparation for 5 minutes, preferably on a heated table, without washing.
9. Wash with distilled water for 3-4 seconds.
10. Cover the tissue section with 10 drops of F- Masson aniline blue solution. After 5 minutes, shake the slide to remove the reagent from the preparation.
11. Wash with distilled water.
12. Dehydrate by passing through the alcoholic stages for a total of less than 1 minute, clean, cover with xylene-based covering medium and examine under a microscope.
NOTE: Consider the initial information leaflet for the staining time. Reduce or increase the duration of each stage differently according to the results obtained in your subsequent studies. At each stage, note the time in the prospectus in which you obtain the most suitable visual result according to your own habits and paint accordingly. |
