| Working Principle
|
The Masson-Fontana staining method is based on the principle that melanin and argentaffin granules react with reducing silver solutions to precipitate metallic silver. Melanin and argentaffin granules reduce silver nitrate and cause the formation of metallic silver. This process produces a black or dark brown color in the tissue. |
| Warnings, Precautions, Recommendations

Danger. Causes severe skin burns and eye damage. IF ON SKIN (or hair): Remove/Take off immediately all contaminated clothing. Rinse skin with water/shower. Wear protective gloves/protective clothing/eye protection/face protection. IF IN EYES: Rinse cautiously with water for several minutes. Remove contact lenses, if present and easy to do. Continue rinsing. |

| Application
|
1. Immerse the preparation in the “Ammonia silver nitrate solution” container. Incubate for 1 hour in the oven at 58-60 0C or place in the microwave at 500 watts for 1 minute. When the time is up, remove by shaking.
2. Wash 3 times with distilled water.
3. Immerse the preparation in “C-Gold chloride solution”. Remove by shaking after 10 minutes.
4. Wash 3 times with distilled water.
5. Immerse in “D-Sodium thiosulfate solution”. Remove by shaking after 5 minutes.
6. Rinse in running tap water for 2 minutes. Then, wash with distilled water 2 times.
7. Cover the preparation with “E- Dark red nucleus solution” for 10 minutes.
8. Rinse in running tap water for 2 minutes. Then, wash with distilled water 2 times.
9. Rinse with 80%, 90% and 100% alcohol.
10. Clean with xylene.
11. Cover with synthetic resin (preferably GulMount).
12. Examine under a microscope. |
