Purpose
Acetocarmine stain is a biological dye used in a technique known as the Belling method. This stain is used specifically for the examination of chromosomes and cellular structures under the microscope. Acetocarmine is a form of carmine dye mixed with acetic acid and is generally preferred for cytogenetic and plant histology studies.
Working Principle
Acetocarmine selectively stains specific cellular structures, especially chromosomes and cell nuclei. Acetocarmine stain is used to visualize chromosomes during mitosis and meiosis. This allows the structure and behavior of chromosomes to be studied.
It is widely used for counting chromosomes and analyzing their structure, especially in plant cells. Used in the study of plant embryos and other cellular structures.
It allows the general morphology of cells and the structure of the nucleus to be characterized.

Application
1. For this purpose, plant spikes are taken and firstly they are subjected to fixation processes. Acetic Alcohol Fixative and Carnoy Fixative should be preferred for fixation. If more successful results are desired, the sample should be left in 70% ethyl alcohol at 0 °C for 1-3 days after fixation.
2. A section is taken from the spike in a tilted position.
3. Place 1 drop of 2809-Acetocarmine Stain, Belling on a clean slide. The dye can be used as it is or diluted twice. The following solution is used for this dilution. Take 1 part of the following mixture and 1 part of 2809-Acetocarmine Dye, Belling dye.
45 ml acetic acid (or propionic acid)
55 ml distilled water
4. As soon as a cross-section is taken from the sample and without waiting, immediately place it on the paint drop.
5. Using the tip of a metal corer, gently press down on the cross-section to ensure that the dye penetrates well into the cross-section.
6. The specimen is removed from the dye drop with the corer.
